Full agonist of the μ-opioid receptor (MOR) with K_i of 383 nM. It also weakly agonizes the δ-opioid receptor (DOR) with a K_i >10 μM. Tianeptine has antidepressant effects but can also regulate neuroplasticity in the amygdala and stress-induced release of glutamate. It can generate analgesia and reward, typical opiate-like behavioural effects, but does not induce tolerance or withdrawal.

Tianeptine has antidepressant and anxiolytic (anti-anxiety) properties with a relative lack of sedative, anticholinergic and cardiovascular adverse effects, thus suggesting it is particularly suitable for use in the elderly and in those following alcohol withdrawal; such persons can be more sensitive to the adverse effects of psychotropic drugs. Recent results indicate possible anticonvulsant (anti-seizure) and analgesic (painkilling) activity of tianeptine via immediate or downstream modulation of adenosine A1 receptors (as the effects could be experimentally blocked by antagonists of this receptor).

Details

Tianeptine Sulfate/Tianeptine hemisulfate monohydrate is an improved salt of Tianeptine. It is not hygroscopic so handling the powder is a lot easier. Since it is not as readily absorbed and excreted from the body, the sulfate salt allows for a more controlled release of the Tianeptine over longer periods. Rather than having to dose three times with rapid peak effects and rapidly falling effects, the sulfate salt allows for one dose to maintain plasma levels in the body much longer than the sodium salt. These improved characteristics make Tianeptine Sulfate a novel addition.
